Preparation, Characterization and Hepatoprotective Potential Activity of Dual Loaded Flavono Polymeric Based Nanoformulation

Abstract

Objective: Flavonoids which are widely spread in plants. This can be categorized as flavonols, flavanols, flavanones, flavones, anthocyanidin and isoflavones. Among all the categories Quercetin, Rutin  and Silibinin majorly used intake in the diet as a functional food. Therefore, flavonoids are better alternative for treating hepatic diseases and its associated complications. However, exploitation of flavonoids Quercetin, Rutin and Silibinin possessing Hepatoprotective activities are hammer, on account of its limited solubility and bioavailability.
 
Methods: For this reason, only in our present study intending to fabricate the dual loaded nano formulation containing Quercetin, Rutin and Silibinin has been performed using biodegradable polymer EPO 100 with poloxamer 407 as surfactant. Nanoprecipitation is the method used for the preparation of nano formulation. Result and conclusion: Prepared nanoformulations were subjected for characterization studies like Molecule interaction by FT-IR , Zeta size, PDI by Malvern , and surface morphology by FESEM and performed In vivo studies acute, sub acute toxicities and hepatoprotective activity. Invivo toxicological studies confirmed that prepared Plain Quercetin, Rutin Silibinin, Qu-Ru and QU-Si nano formulations does not produce any sign of toxicity at given single and multiple oral dose and found to be safe for clinical utility The nanonized dual loaded Quercetin-Rutin (Qu-Ru NPs), Quercetin-Silibinin (Qu-Si NPs) Polymeric nano formulation displayed one fold enhanced hepato protective activity against paracetamol intoxicated in comparison with pure compound and single loaded polymeric nano formulation.
