Genetic Polymorphism in Eucalyptus tereticornis Sm. Using RAPD Markers

Abstract

In India Eucalyptus is grown in over 8 million hectare area with an average productivity to about 5-7 m3ha-1yr-1. However, it is considered to be a stable species with resistance to genetic breakdown and possibility of higher productivity even under un-irrigated conditions to about 20-25 m3ha-1yr-1. In recent past, productivity of species has been increased by application of genetic tools and clonal approach to multiply and commercially deploy genotypes with promising traits. The present investigations purports towards screening genotypes that were not only productive and stable but genetically divergent.  Genetic diversity in twenty five genotypes of Eucalyptus tereticornis Sm. was analyzed using randomly amplified polymorphic DNA (RAPD) with 10 decamer primers. The number of scorable bands for each primer varied from 6 (OPA-2) to 14 (M-131) with an average of 10 bands per primers. A total of 96 distinct DNA fragments (bands) were amplified, of which 80 bands were polymorphic ranged from 200 to 1300 base pair. In fact the number of amplified bands per genotypes varied from 5 to 13 and polymorphism percentage ranged from 73.00 to 93.00 with an average of 83.32. The mean polymorphic information content (PIC) for RAPD primers was recorded to be 0.34 which ranged from 0.19 to 0.44. The Jaccard similarity coefficient ranged from 0.16 to 0.90 showing wide range of variability among the genotypes. Interestingly, most of the clones which originated from same provenance tend to group in a single cluster. However, clones from same location dispersed well in different clusters as well.
