Mahendra K. Singh

Designation: Research Assistant Professor University: University of Miami Country: USA More Details
Currently, our research is focused on deciphering the role of a multi-domain scaffolding protein NEDD9 in gastrointestinal cancers and breast cancer. NEDD9 provides a signaling complex regulating multiple cellular processes relevant to cancer. These include responsiveness to signals emanating from the T and B cell receptors, integrins, chemokine receptors, and receptor tyrosine kinases, as well as cytoplasmic oncogenes such as BCR-ABL and FAK- and SRC-family kinases. NEDD9 is known to influence functions as migration, invasion, survival, and mitosis.

We discovered a novel protein CASS4 which is currently being studied for its role in regulation of metastasis and immune components in lung cancers.

In addition, we are identifying molecular subtypes in gallbladder cancer with the goal of characterizing new alterations, confirming genetic differences in these distinct subtypes of gallbladder cancer so as to decipher potentially targetable genetic aberrations.

Natural products (NPs) are pivotal resources for the discovery of bioactive entities. They harbor unique structural diversity, novelty, three-dimensionality, and complexity. Thus, imparting them uniqueness for interrogating complex biological systems. NPs generously offer versatile bioactive scaffolds that are tractable to rationally-designed semisynthetic transformations for exploring structure-activity relationship (SAR), improving biological potency, enhancing target selectivity, and reducing associated undesirable effects.
Protein kinases orchestrate multiple signaling pathways of the living cells and their dysregulation is frequently associated with various diseases, including cancers. Consequently, extensive efforts have been devoted for the discovery and development of novel candidates that can modulate the catalytic activity of protein kinases and so the revoke disease pathogenesis. Kinases inhibitors are currently playing an increasingly notable role in cancer treatment. To date, more than 27 oncology drugs, which target kinases, have been approved for marketing and numerous additional such therapeutics are in various stages of clinical evaluation. This truly emphasizes the eminent role of kinases as unique druggable targets.

Research Interests: Natural products; Rational design; Semisynthetic transformation, Cancer, Protein kinases.
